Come to think of it, the Mines pass through the lower part of Darkness's house (we see a track underneath the area with the baskets, which is much lower than the kitchen and much lower than the lab would be), so the door to the Mines should not be at that level either. He is now level with the kitchen and to the right of it, thus, he shouldn't be able to go straight up into the kitchen via the dumbwaiter. However, to get to the laboratory, Sam has to go up the stairs (which is 'up' and 'back,' if one is looking at Darkness's entrance hall from the angle that it is shown in the game), through the Doors of Knowledge (which, relative to the gravity of Darkness's house outside the Doors of Knowledge, means he's going further to the right), further upward and 'forwards' (assuming the den and the room outside the Doors of Knowledge have the same gravity relative to one another), then down a flight of stairs that take him to a level that, judging by the length and angle of the flight of stairs, is probably level with the Doors of Knowledge and finally, down another flight of stairs into the lab.
